- <b>ABSTRAK</b><br> Kanker serviks merupakan kanker peringkat kedua yang paling umum menyerang wanita dan disebabkan oleh infeksi human papillomavirus. Ekspresi protein E6 dan E7 human papillomavirus tipe risiko tinggi dapat menginduksi proses pembentukan tumor (Tumorigenesis). Penelitian bertujuan untuk mengekspresikan protein fusi E6/GFP dan E7/GFP pada sel HeLa. Plasmid pCDNA 3.1 yang membawa gen E6 dan E7 HPV-16 yang telah dimodifikasi, serta GFP sebagai reporter gene ditransfeksi ke dalam sel HeLa menggunakan metode elektroporasi. Hasil elektroporasi dianalisis menggunakan mikroskop konfokal. Hasil elektroporasi memperlihatkan perpendaran hijau pada sel yang dielektroporasi dengan gen E6 dan E7. Hasil penelitian menunjukkan protein fusi E6/GFP dan E7/GFP telah berhasil diekspresikan pada sel HeLa dengan persentase transfektan E6/GFP sebesar 0,0091% lebih tinggi dibandingkan E7/GFP sebesar 0,0002% maupun GFP sebesar 0,0022%. <hr> <b>Abstract</b><br> Cervical cancer ranked second as the most common cancer affecting women that is caused by human papillomavirus infection. E6 and E7 proteins expression of human papillomavirus can induce the process of tumor formation (tumorigenesis). The study is aimed at expressing the E6/GFP and E7/GFP fusion proteins in HeLa cells. Plasmid pCDNA 3.1 that carry modified E6 and E7 genes of HPV-16, as well as the GFP reporter gene was transfected into HeLa cells by electroporation method. The results of electroporation was analyzed by confocal microscopy. The results showed the green fluorescence was observed in cells that were transfected with E6 and E7. The results confirmed that the E6/GFP and E7/GFP fusion proteins were successfully expressed in HeLa cells with higher level of expression E6/GFP is 0,0091% in comparison with E7/GFP is 0,0002% or GFP 0,0022%.;
