Kemampuan suatu mikrosfer untuk melepaskan agen farmasetika bergantung pada kemampuannya untuk terdegradasi. Keseragaman ukuran partikel mikrosfer, serta komposisi polipaduan akan mempengaruhi degradasi mikrosfer polipaduan. Dilakukan uji degradasi terhadap mikrosfer polipaduan secara in vitro dengan menginkubasi mikrosfer pada suhu 37&amp;#730;C di dalam buffer fosfat pH 7,4 selama 8 minggu. Karakterisasi dilakukan terhadap penentuan persen kehilangan berat, pengamatan bentuk dan morfologi dengan mikroskop optik dan SEM. Persen kehilangan berat semakin meningkat seiring dengan lamanya waktu inkubasi. Mikrosfer pada inkubasi minggu ke-1 diperoleh rentang persen kehilangan berat sebesar 8,8 &#x2013; 12,5 %, sedangkan pada minggu ke-8 diperoleh rentang persen kehilangan berat sebesar 20,6 &#x2013; 25,7%. Semakin besar komposisi PLA dalam polipaduan, persen kehilangan berat mikrosfer cenderung meningkat pada beberapa komposisi. , Polyblend of poly (lactic acid) (PLA) and polycaprolactone (PCL) microspheres could be prepared as a pharmaceutical coating agent for controlled release purposes. The ability of microspheres to release the pharmaceutical agent depends on its ability to degraded. Homogenity of particle size of microspheres and polyblend composition will affect the degradation of polyblend microspheres. In vitro degradation of polyblend microspheres was done by incubating microspheres at 37&amp;#730;C in phosphate buffer pH 7,4 for 8 weeks. Characterization to determine the percent loss of weight, shape and morphology observation with optical microscope and SEM. Percent weight loss increased along with the length of incubation time. Microspheres at 1st week incubation has a percent weight loss th range from 8,8 to 12,5%, while at 8week has a percent weight loss range from 20,6 to 25,7%.
