Karakteristik Kolagen Kulit Tuna Sirip Kuning (Thunnus albacares) pada Waktu Hidro-Ekstraksi Berbeda dan Potensinya dalam Bentuk Sediaan Nanokolagen

  • This study aimed to analyze the effect of hydro-extraction time on the characteristics of yellow fin tuna skin collagen and its potential in the form of nano-collagen preparations. This study was designed using a Completely Randomized Design (CRD). The treatment factor was the length of time for hydro-extraction of collagen which consisted of three levels, such as 1 hour, 3 hour, and 5 hour with two repetitions. The research data were analyzed by analysis of variance (ANOVA) and Duncan Multiple Rangeâ€TMs Test. The results showed that the hydro-extraction time had a significant effect (p<0,05) on the yield, protein content, and fat content. The best treatment was 5 hours hydro-extraction time with 4.6% yield, 82.96% of protein content, 1.91% of moisture content, 0.74% of fat content and pH value of 5.52. The results of the nano-collagen particle size observation based on the best treatment ranged from 73.80–693.72 nm.Keyword:       Hydro-extraction, nano-collagen, particle size, tuna skin, characteristics Penelitian ini bertujuan untuk menganalisis pengaruh lama waktu hidro-ekstraksi terhadap karakteristik kolagen kulit ikan tuna sirip kuning dan potensinya dalam bentuk sediaan nano-kolagen. Penelitian ini dirancang menggunakan metode Rancangan Acak Lengkap (RAL). Faktor perlakuan adalah lama waktu hidro-ekstraksi kolagen yang terdiri dari 3 level, yaitu 1, 3 dan 5 jam dengan dua kali pengulangan. Data hasil penelitian yang diperoleh dianalisis dengan analisis ragam (ANOVA) dan uji lanjut Duncan. Hasil analisis sidik ragam menunjukkan lama waktu hidro-ekstraksi berpengaruh nyata (p<0,05) terhadap rendemen, kadar protein dan kadar lemak kolagen. Perlakuan terbaik yaitu lama waktu hidro-ekstraksi 5 jam dengan persentase rendemen 4,69 %, kadar protein 82,96 %, kadar air 1,91 %, kadar lemak 0,74 % dan nilai pH 5,52. Hasil pengamatan ukuran partikel nano-kolagen yang diperoleh berdasarkan kolagen perlakuan terbaik berkisar antara 73,80–693,72 nm.Kata kunci:   Hidro-ekstraksi, nano-kolagen, ukuran partikel. kulit tuna, karakteristik
