- Background: Increasing evidence suggests that Alzheimer’s disease (AD) pathogenesis and progression are not only restricted to the neuronal compartment, but also involve changes in distribution and activation of specific immune cell populations. Given the critical involvement of the endocannabinoid system (ECS) in modulating the activation and responses of all immune cells, we asked whether a possible correlation between the molecular changes of fatty acid amide hydrolase (FAAH), the main degrading enzyme of endocannabinoids, exists in several peripheral blood immune cells in AD patients. Results: We analyzed the expression of FAAH in CD3+ T-lymphocytes, CD19+ B-lymphocytes, CD14+ monocytes and CD56+ NK cells obtained from peripheral blood of healthy donors and AD patients with increasing cognitive impairment by polychromatic flow cytometry. We found that only monocytes showed a significant increase of FAAH in AD patients, while its levels remained unchanged in the other immune cell subsets (Fig.1). Furthermore, the expressions levels of FAAH negatively correlated with disease severity of AD patients, expressed as MMSE, inasmuch as monocytes obtained from the most severe subjects had the highest FAAH expression (Fig.2). Conclusions: Taken together, our findings not only are instrumental to better characterize the ECS within the immune system but will possibly lead to novel monocyte-centered therapeutic approaches aimed at shifting them towards and anti-inflammatory and neuroprotective profile.
