Identifying Potential New Stem Cell OCT4 Biomarker in Breast Cancer

- For breast cancer diagnosis and prognosis, only a few biomarkers have been identified. However, no precise biomarkers exist that can discriminate between aggressive breast tumours that require drastic treatment and those with a good prognosis. As a result, finding novel breast cancer biomarkers has become a top focus. Potential stem cells found within tumours have received a lot of attention recently, and it's thought that these cells have a role in cancer development, progression, and therapy resistance. The aims of this study were to focus on proteins linked to stem cells, cellular differentiation and/or tumour formation and progression and establish if they are differentially expressed between normal and malignant breast tissues and/or between samples with different clinical features such as grades and stages. The study was conducted in the two main hospitals The-Hussein teaching hospital and Al-haboubi hospital (cancer unit) in Thi-Qar province during the period from October 2020 to May 2021. The project include63 breast tissue samples divided into two groups: The breast cancer group consists of 53 tissue samples and control group consists of 10 benign breast tissues. The age range was between 20-60 years old. Expression of the Oct3/4 protein in tumors was confirmed by immune histochemistry staining. A potential biomarker was identified using literature searching. The Oct4 expression was evaluated by immunohistochemistry using 63 breast tissue samples. This study found that nuclear and cytoplasmic staining for Oct4in both benign and malignant breast tissues. Nuclear Oct4 staining was increased significantly in breast cancer and negatively associated with increasing grade and positivity associated with tumour size. In contrast, cytoplasmic Oct4 staining was not associated with breast cancer and its clinical features such as grades and stages.
