- Background: Shigella flexneri is the most common bacteria causeddiarrhea with number of cases increased each year. Mangosteen peel hasbeen used for years by most of Indonesian as a traditional medicine for thetreatment of diarrhea, dysentery, and infected wound. Experimental studieshave demonstrated that extract of Garcinia mangostana L. peel hasantimicrobial activity. Objective: This study aimed to investigate theantibacterial activity of ethyl acetate fraction of Garcinia mangostana L. peelmethanol extract and the effective concentration to inhibit the growth ofShigella flexneri. Methods: The ethyl acetate fraction obtained from liquidfractionation of methanolic extract. Extracts were phytochemically testedand antibacterial activity test by well diffusion method against Shigellaflexneri. Positive control used 5μg ciprofloxacin and negative control used10% DMSO. Results: Phytochemical screening of ethyl acetate fraction ofGarcinia mangostana L. methanol extract contained alkaloid, saponin,tannin, flavonoid, and triterpenoid. The effective concentration to inhibit thegrowth of the Shigella flexneri was 10% (p = 0.000). Conclusion: Ethylacetate fraction of methanol extract of Garcinia mangostana L. peel hasantibacterial activity against Shigella flexneri.Keywords: Antibacterial, ethyl acetate fraction of Garcinia mangostana L.methanol extract, Shigella flexneri
