FORMULASI SEDIAAN TABLET ANTIOKSIDAN DARI EKSTRAK KULIT KAYU SECANG (Caesalpinia sappan L.) DENGAN METODE KEMPA LANGSUNG

- Telah dilakukan penelitian mengenai teknologi dan formulasi pembuatan tablet dari ekstrak secang (Caesalpinia sapppan L.). Penelitian ini bertujuan untuk membuat sediaan tablet ekstrak secang yang memenuhi persyaratan sebagai suatu sediaan farmasi. Pada penelitian ini telah dibuat empat formula menggunakan metode kempa langsung. Masing-masing formula mengandung ekstrak secang sebagai zat aktif, FUJICALIN dan pati jagung sebagai pengisi, magnesium stearat dan talkum sebagai pelincir, AEROSIL 200 sebagai absorben. Evaluasi massa cetak tablet yang diuji meliputi kadar air, kecepatan alir dan sudut istirahat, kerapatan nyata, kerapatan mampat, kompresibilitas, kerapatan sejati. Evaluasi tablet yang diuji meliputi keseragaman bobot, keseragaman ukuran, kekerasan, kerapuhan, waktu hancur, kromatografi lapis tipis. Pengujian aktivitas antioksidan tablet dilakukan dengan metode Diphenylpicrylhydrazyl (DPPH). Metode analisis statistik yang digunakan adalah analisis varian dengan uji lanjut Newman-Keuls dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa semua formula memenuhi persyaratan sebagai suatu sediaan farmasi menurut Farmakope Indonesia IV. Formula yang tepat untuk keempat formula yaitu menggunakan konsentrasi FUJICALIN sebagai zat pengikat lebih dari 55%. Hasil analisis kromatografi lapis tipis menunjukkan bahwa komponen metabolit sekunder pada ekstrak masih terdeteksi pada sediaan tablet. Hasil pengujian antioksidan terhadap keempat formula tablet ekstrak tergolong dalam kategori antioksidan sangat kuat karena nilai IC50 kurang dari 50 ppm.The research has been done about technology and formulation of production the tablet from extract secang (Caesalpinia sapppan L.) The research is aimed for making preparation tablet extract secang that is fulfilled the requirement as a pharmaceutical preparation. In this research, there has been made four formulas using a direct compress method. Each formula contains extract secang as active substance FUJICALIN and corn starch as filler, magnesium stearate and talc as lubricant, AEROSIL 200 as absorbent. The evaluation of print mass tablet that is checked comprises water levels, flow velocity, angle of rest, real density, incompressible density, compressibility, true density. The evaluation of tablet that is checked involves uniformity of size, hardness, friability, disintegration time, thin layer chromatography.The test of antioxidant activity tablet is done with Diphenylpicrylhydrazyl (DPPH) method. Analysis statistics method that is used is analysis of variance with further test Newman-Keuls with 95% confidence level. The result of research shows that all formulas fill the requirements as a a pharmaceutical preparation depends on Indonesian Pharmacopoeia IV. The appropriate formula for four formulas uses FUJICALIN concentrate as binder substance more than 55%. The result of analysis thin-layer chromatography shows that component of secondary metabolites in the extract is still detected in tablet preparation.The result of antioxidant test show the highest activity in formula IV because contains IC50 9,88 ppm.Keywords: Tablet, Direct compress, Extract, Secang, Antioxidants.
