  • ABSTRAKLisdiyariniN.2017.ProduksidanPemekatanKeratinaseBacillussp.MD24sertaKemampuannyasebagaiAgenDehairingKulitKambing.SkripsiJurusanKimiaFMIPAUniversitasNegeriMalang.Pembimbing(I)Dr.SuhartiS.Pd.M.Si.(II)MuntholibS.Pd.M.Si.KataKunciBacilluskeratinasedehairingPemanfaatankulitkambingdalamindustripenyamakankulitterusmeningkat.Namundalamsalahsatutahapannyaterdapatprosesdehairing(buangrambut)yangberkontribusimenghasilkanlimbahberbahayaberupagasH2Sdanlumpurkapur.Berbagaiupayadikembangkanolehparapenelitiuntukmeminimalisasibahayadarilimbahprosespengolahankulittersebutsalahsatunyadenganpemanfaatanenzimkeratinase.Enzimkeratinasemerupakanenzimproteasespesifikyangmampumendegradasikeratin.KeratinaseyangdihasilkanolehbakteriBacillussp.MD24mampumendegradasibuluayamdanmerontokkanrambutpadakulitkambingdalamwaktuyanglama(72jam).Lamanyawaktudehairingdapatdisebabkanolehrendahnyaaktivitasekstrakkasarkeratinase.Peningkatanaktivitasenzimdapatdilakukandenganmengoptimalkanproduksienzimdanmelakukanpemekatanekstrakkasarkeratinase.Penelitianinibertujuanmeningkatkankemampuanenzimkeratinasesebagaiagendehairingkulitkambingdengan(1)mengetahuipengaruhpenambahan05%dan1%sumberkarbontambahan(sukrosaglukosadanfruktosa)terhadapproduksiekstrakkasarkeratinase(2)mengetahuipengaruhwaktupenyimpananterhadapaktivitaskeratinase(3)mengetahuikemampuanenzimkeratinasehasilpemekatanmenggunakanamoniumsulfat80%jenuhdalamprosesdehairingkulitkambing.Tahap-tahappenelitianiniterdiridari(1)peremajaanbiakanmurnibakteriBacillussp.MD24(2)produksiekstrakkasarkeratinaseBacillussp.MD24(3)ujiaktivitaskeratinaseBacillussp.MD24(4)pengaruhkonsentrasisumberkarbontambahan(sukrosaglukosadanfruktosa)danwaktuinkubasiterhadapproduksikeratinase(5)pengaruhwaktupenyimpananterhadapaktivitasekstrakkasarkeratinase(6)pemekatanekstrakkasarkeratinasedenganamoniumsulfat80%dan(7)ujikemampuankeratinaseBacillussp.MD24sebelumdansesudahpemekatansebagaiagendehairingkulitkambing.KondisiterbaikatauoptimumdiketahuiberdasarkannilaiaktivitaskeratinasedandidukungolehnilaiWeightLoss.Hasilpenelitianinimenunjukkanbahwa(1)penambahan05%dan1%sumberkarbontambahan(sukrosaglukosadanfruktosa)menurunkanproduksiekstrakkasarkeratinase(2)Semakinlamawaktupenyimpananekstrakkasarkeratinasemakaaktivitaskeratinasemengalamipenurunansecarabertahapdan(3)Enzimkeratinasehasilpemekatanmenggunakanamoniumsulfat80%jenuhmemilikiaktivitaskeratinaseyangsebelumnyayaitu8793U/mLmenjadi20288U/mLdanmampusebagaiagendehairingkulitkambingpadaperendamanselama48jam.ABSTRACTLisdiyariniN.2017.TheProductionandPreconcentrationofBacillussp.MD24KeratinaseanditsAbilityasGoatSkinDehairingAgent.UndergraduateChemistryDepartmentMathematicsandScienceFacultyStateUniversityofMalang.Advisors(I)Dr.SuhartiS.Pd.M.Si.(II)MuntholibS.Pd.M.Si.KeywordsBacilluskeratinasedehairingTheutilizationofgoatskininthetanneryindustrycontinuestoincrease.HoweverinoneofthetanningstagesthereisadehairingprocessthatcontributestotheproductionofhazardouswasteintheformofH2Sgasandlimemud.Therearevariouseffortsdevelopedbyresearcherstominimizethehazardsofthewaste.Oneofthemethodsisbyutilizingkeratinaseenzyme.Keratinaseenzymeisaspecificproteaseenzymecapableofdegradingkeratin.KeratinaseproducedbyBacillussp.MD24especiallyisabletodegradechickenfeathersandpromotehairlossongoatskinforalongtime(72hours).Thelengthofthedehairingtimecouldbecausedbythelowactivityofthecrudeextractofkeratinase.Enzymeactivitymightbeincreasedthroughoptimizationoftheproductionofenzymesandpreconcentratiionofthekeratinasecrudeextract.Hencethisstudyaimstoimprovetheabilityofkeratinaseenzymeasagoatskindehairingagentby(1)knowingtheeffectofadding0.5%and1%additionalcarbonsource(sucroseglucoseandfructose)totheproductionofcrudekeratinaseextract(2)knowingtheeffectofstoragetimeonkeratinaseactivity(3)knowingthecapabilityofconcentratedkeratinaseenzymebyusing80%saturatedammoniumsulphateingoatskindehairingprocess.Thereaseachwasdonein7steps(1)regeneratingthepurecultureofBacillussp.MD24(2)producingthecrudekeratinaseextractfromBacillussp.MD24(3)determiningthecrudeextractkeratinaseactivityfromBacillussp.MD24(4)observingtheeffectoftheconcentrationofadditionalcarbonsources(sucroseglucoseandfructose)andincubationtimeonkeratinaseproduction(5)observingtheeffectofstoragetimeontheactivityofcrudekeratinaseextract(6)concentratingthecrudeextractofkeratinasewith80%ammoniumsulphateand(7)testingdancomparingtheabilityofkeratinaseBacillussp.MD24beforeandafterpreconcentrationasagoatskindehairingagent.ThebestoroptimumproductionconditionsaredeterminedbasedonthevalueofkeratinaseactivityandsupportedbythevalueofWeightLoss.Theresultsofthisstudyindicatedthat(1)theadditionof0.5%and1%additionalcarbonsource(sucroseglucoseandfructose)decreasedtheproductionofcrudekeratinaseextract(2)thelongerthestoragetimeofcrudeextractskeratinasethelesserkeratinaseactivityand(3)thepreconcentratedkeratinaseenzymeusing80%saturatedammoniumsulphatehasahigherkeratinaseactivityfromtheearlierstate8793U/mLto20288U/mLanditcapableofactingasagoatskindehairingagentforimmersionfor48hours.
