  • Red roselle (Hibiscus sabdariffa L.) calyx which contains flavonoid allegedly as potential teratogenic agent. General toxicity test that has been done previously such as acute and subchronic toxicity test obtained LD50 value > 15000 mg/ kgBW that can be categorized as non toxic and safe to use. Specific toxicity test such as teratogenic test is does to complete the safety data of red roselle plant and to discover the effect on embryo if consumed by pregnant woman. The doses administered were 20.5, 41, and 82 mg/20 gBW. The extract was administered during the organogenesis days of 9th to 17th day of pregnancy, and the pregnant mice were dissected on 18th day to observed fetus weight, life fetus, dead fetus, implantation, resorption, fetus external abnormalities, and fetus skeletal disorders. The result of statistic analysis with ANOVA showed that extract had significant effect on pregnant mice weight gain, fetus weight and inhibition of sternum forming (asymmetric shaped) (F > F crit 0.05) and very significant effect on inhibition of interparietal, supraoccipital, sacrocaudal, sternum (asymmetric cleaved and asymmetric dumbbell shaped), anterior intermediate phalanx, posterior proximal phalanx, and posterior intermediate phalanx (F > F crit 0.01). Duncan multiple range test showed P1 dose (20.5 mg/20 gBW) was the minimum dose of teratogenic effect (P1 > DMRT 0.05), it can be concluded that ethanolic extract of red roselle has teratogenic effect on mice (Mus musculus) fetus.
