UJI ANTIHIPERGLIKEMIK EKSTRAK ETANOL BIJI MELINJO (Gnetum gnemon L.) TERHADAP TIKUS PUTIH JANTAN GALUR WISTAR YANG DIINDUKSI ALOKSAN DAN HISTOPATOLOGI PANKREAS

- Melinjo seeds (Gnetum gnemon L.) have activities as antihyperglycemic. This study was conducted to determine the effect of giving ethanol extract of melinjo seed (EEBM) toward the reduction of blood glucose levels in Wistar strain male rats induced by alloxan and pancreatic histopathology. Phytochemical screening showed that EEBM contained alkaloid compounds, flavonoids, polyphenols, and saponins. EEBM used has a level of ethanol and water-soluble extract of 70.70 ± 0.26 and 46.20 ± 0.25, water content and total ash content of 6.40 ± 0.54 and 7.53 ± 0.28. EEBM was given to mice with variations in the dosages of 125, 250 and 500 mg/kgBB. As a comparison of normal control without treatment, positive control of 1 IU/kgBB insulin and negative control of 0.5% Na-CMC suspension. Tests were carried out by measuring blood glucose levels of mice by GOD-PAP method using DTN-410-K photometer on days 0, 5, 10, 15, and 20 and followed by histopathological examination of the pancreas by hematoxylin-eosin staining method. The average percentage results of blood glucose levels for the positive control group is 40%, and the three treatment groups are 36%, 40%, and 44% respectively. This shows that the treatment of a dose of 500 mg/kgBB had the highest antihyperglycemic activity compared to the other 2 treatments and there were significant differences (p<0.05) compared to insulin. Histopathological features showed nearly perfect improvement in endocrine cells of the Langerhans island at doses II, III and positive controls compared to negative controls and doses I. ED50 value EEBM is 767.45 mg kgBB. The conclusion of this study is that EEBM has low antihyperglycemic activity.
