Molecular Variation of Oil Palm (Elaeis guinnensis Jacq.) Clones Based on RAPD (Random Amplified Polymorphic DNA) Marker

  • The objective of this research was to analyze molecular variation of Oil Palm (Elaeis guinnensis Jacq.) BTC-60 clone based on RAPD used marker OPD-13, OPD-20 and OPM-01. This research was conduted at Plant Tissue Culture Laboratory, Faculty of Agriculture, University of Sumatera Utara, Medan, from March 2017 until June 2017. Genetic materials were 30 DNA stock of oil palm BTC-60 clone from PT. Socfindo. A total 13 bands were scored, with the range of band size was 405 bp – 2268 bp and average of polymorphism of primers was 39,29%. Genetic distance measurement and dendrogram formation were analyzed using DARwin 6.0 software. Based on the analysis, 30 clones of oil palm grouped into 3 clusters with a total of molecular diversity was 65,89%. The range of dissimilarity coefficient was 0-0,24. OPD-13 marker showed the presence of specific bands, 5th band (634 bp 20, 22 clone; 6th band (557 bp) in 16, 22 and 28 clone; and 7th band (461 bp) in 2, 16, 22 and 28 clone.
