  • Amorphophallus variabilisBlume, a member of Araceae, is a native tuber crop in Java, Madura and Kangean Islands, Indonesia. The plant showed high variations in morphology. However, genetic variations at molecular level have not been well studied. Amplified fragment length polymorphism (AFLP) was carried out using 8 primers combination of EcoRI and MseI on 78 accessions collected from 28 sites in Java, Indonesia. Results showed that AFLP markers able to generate polymorphism among accessions. A total of 220 polymorphisms were found. The differences among accessions at the genetic level were high, and 5 clusters were constructed. Grouping was independent of geographical origin, similar to clustering of morphological characteristic of flowers as in the previous report. Accessions from one site composed of one to four different cluster groups, showed that variation in single site was observed. Regarding conservation program of the A. variabilisin natural population, it is reasonable to protect one bigger site rather than many small sites, but it should be recommended to maintain conservation areas in several districts. Further study on population structure should be carried out to explain such variability. Keywords: genetic diversity, iles-iles, molecular marker, morphology, tuber crop

  • The objective of this study was to evaluate the characteristics of mangosteen fruit’s hull, xanthone content, and antioxidant potential on various stadia of mangosteen fruit development. The experiment was conducted in September 2006 until July 2008 using randomized complete block design, with three replications at maturation stage i.e. 1, 2, 3, and 4 months after anthesis. The parameters being observed were fruit’s hull characters, xanthone content, and antioxidant capacity. The results of this study showed that fruit diameter and fruit weight increased until three months after anthesis and then did not change significantly once they entered the process of maturity. Thickness of fruit’s hull differed significantly among maturation stage. The thickness of fruit hull was observed at two months and the highest weight was at three months after anthesis. Xanthone content of mangosteen fruit’s hull at a month up to four months after anthesis did not differ significantly however capacity of antioxidant differed significantly among fruit ages, the IC50 values increased with the increase of fruit maturation. Keywords: Antioxidant, fruit growth, fruit’s hull, mangosteen, xanthone

  • One method to obtain genetic information is the diallel cross analysis. The objective of this study was to eavluate the genetic parameters of six inbred pepper (Capsicum annuum L.) using full diallel crosses. The experiment was conducted at IPB Experiment Field, Cikabayan, Darmaga. The design was randomized complete block design (RCBD) using three replications as blocks. Data from generation F1 and parents were analyzed using the Hayman Method. Results indicated that no epistatic effects were significant for all the traits assessed. Additive genetic effects were larger than the dominant effects for yield per plant, fruit length, and diameter fruit traits. Dominant genetic effects were larger than the additive effects for fruit weight traits. Narrow-sense and broad-sense heritability were high for all the traits assessed. The character of the yield per plant, fruit weight and fruit diameter shows that there were more dominant genes in the parents. There were more recessive genes in parents for the fruit length character. IPB C7 parent was the most recessive genes containing control characters in the yield per plant. In the new improved varieties of high yielding, IPB C7 could be crossed with IPB C9. Employing individual or mass selection breeding should be successful in developing high-productivity lines in this population
